Gabapentin (GBP) is superior to pregabalin (PGB) in the treatment of patients with chronic sciatica (CS), with fewer and less severe adverse events (AEs), according to a study published October 15 in JAMA Neurology.

Kelvin Robertson and colleagues from Townsville Hospital in Australia conducted an interim analysis of a randomized, double-dummy, double-blind crossover study of PGB and PGB for the treatment of CS; 20 patients were randomly assigned to the PGB and PGB groups, and each received PGB for 8 weeks.

The researchers found that PGB was superior to PHB, with fewer and less severe reported AEs; both PGB and PHB showed a significant reduction in pain intensity on a visual analog scale (PGB: mean 7.54 to 5.82; PHB: mean 7.33 to 6.38) and a reduction in pain intensity on a visual analog scale. 6.38) and a decrease in the Oswestry Disability Index (mean from 59.22 to 48.54). The reduction in the Oswestry Disability Index did not differ between PGB and PGB. Adverse events were more frequent with GHB (81% vs. 19%), especially when GHB was administered first.

GEB was better at reducing pain intensity than PGB, with fewer and milder adverse events.” Gabapentin should be started before GHB to ensure optimal crossover.”
